Two switches, one big pulse

GHRH works through cAMP and the ghrelin receptor works through calcium. When both are pressed at once, the growth hormone pulse is several times larger than either alone. That is why ghrelin-type peptides are almost always paired with a GHRH peptide. The pairing also overrides part of the somatostatin brake that grows with age.
